Schon vor Jahren stellte sich heraus, dass die Zellen des Ewing-Sarkoms einen Todesrezeptor besitzen, der auf das Molekül TRAIL reagiert. Experimente, die Kontny und seine Mitarbeiter an den Krebszellen im Reagenzglas durchführten, zeigten: TRAIL treibt rund 80 Prozent der Tumorzellen in den Suizid. Im Gegensatz zu dem anderen bekannten Molekül Fas Ligand, welches ebenfalls Todesrezeptoren einschaltet, hat TRAIL keine so großen Nebenwirkungen auf andere Gewebe im Körper wie etwa die Leber. Es scheint gezielter gegen Tumorzellen einsetzbar zu sein. „Das macht dieses Molekül natürlich sehr attraktiv für die Krebstherapieforschung“, sagt Kontny. Es könnte schon bald als Bestandteil einer kombinierten Chemotherapie gegen das Ewing-Sarkom eingesetzt werden. Aber warum wirkt TRAIL nur bei 80 Prozent der Tumorzellen? Das untersuchten Kontny und Co. in einem zweiten Schritt. Sie stellten fest, dass bei einem Teil der Krebszellen der Signalweg, der dem Todesrezeptor nachgeschaltet ist, gestört ist. Genau genommen fehlt diesen gegen TRAIL resistenten Tumorzellen das Abbauenzym Caspase 8. Lässt sich dieses Problem beheben?
