De ziekte van Alzheimer (AD) is de meest voorkomende vorm van dementie. Dankzij recente internationale genoomwijde associatie studies zijn 10 nieuwe genen ontdekt die geassocieerd zijn met het risico op het verkrijgen van AD (CLU, CR1, PICALM, BIN1, ABCA7, CD2AP, CD33, EPHA1, MS4A4E en MS4A6A). Het hoofddoel van dit project is om nieuwe genetische risicofactoren te identificeren die een groot deel van de ontbrekende erfelijkheid van AD zouden kunnen verklaren. Om deze volledige nieuwe AD genen, inclusief de up- en downstream regulatorische gebieden, te kunnen onderzoeken in onze Vlaams-Belgische studiepopulatie, zullen we gebruik maken van de in-house next-generation sequencing technologie: SOLIDTM5500xl. De genetische follow-up van deze nieuwe AD genen bestaat uit 5 fasen: Identificatie van een vaakvoorkomende genetische variant die de waargenomen associatie tussen het gen en AD verklaart. Onderzoek naar de aanwezigheid van zowel zeldzame als structurele varianten in de nieuwe AD genen. Als gevolg van de complexiteit van AD zullen we ook genetische interacties onderzoeken tussen de nieuwe AD genen. Verder zullen we genotype-fenotype correlatiestudies uitvoeren met fenotypes variërend van biomerkerprofielen in CSF, plasma of serum tot klinische fenotypes. Tot slot zullen we de geassocieerde variaties functioneel onderzoeken.

Il propose, spécifiquement, des principes permettant de déterminer quelles revendications sont inacceptables, lesquelles sont acceptables, et lesquelles ont, au titre de la justice, un caractère impératif. C’est l’application pratique de tels principes à des cas particuliers qui est appelée ici adjudication, qu’elle ait ou non un caractère strictement judiciaire. L’article défend l’idée que, pour avoir envisagé l’adjudication dans une perspective exclusivement normative, nombre de contributeurs à la réflexion multiculturaliste, y compris certains des plus influents, ont mal situé le problème, et donc proposé des solutions de pertinence discutable. Cet accent normatif résulte du souci compréhensible d’éviter que la justice se confonde avec l’équilibre des intérêts dans un processus pluraliste de marchandage. On y perd toutefois l’épaisseur de la sociologie politique des revendications ethniques, elle-même étroitement liée à l’épaisseur institutionnelle de leur adjudication. Une dimension essentielle de cette perte est la conception sociologiquement insatisfaisante de la culture qui est caractéristique du multiculturalisme normatif, et qui en rend l’application malaisée précisément dans les contextes mêmes dont les théoriciens multiculturalistes se préoccupent. La recherche de principes d’adjudication des revendications ethniques qui ne soient ni purement formels ni assujettis aux dynamiques politiques pratiques, y compris les rapports de forces empiriques, se révèle en fin de compte infructueuse.

Il lavoro dei ricercatori INBB ha già contribuito al settore in maniera significativa dimostrando alta competitività in ambito internazionale. In particolare, le ricerche della UdR di Parma hanno già portato alla messa a punto di approcci terapeutici innovativi basati soprattutto sull’uso di sostanze naturali a scopo chemio-preventivo per il cancro della prostata. I risultati hanno riguardato l’individuazione di sostanze attive, la messa a punto di bio-marcatori utili per il monitoraggio anche personalizzato della risposta terapeutica, l’efficacia clinica della chemio-prevenzione, lo studio della progressione neoplastica e della resistenza alla terapia. Il Cancro della Prostata (PCa) è il secondo più comune tumore maschile dei paesi occidentali, ma diventerà presto il primo a causa dell’aumento dell’età media della popolazione. La mortalità specifica per questa malattia è solo il 15% circa, perché solo la forma metastatica uccide il paziente. La grande maggioranza dei PCa risulta di fatto di natura indolente. Ma il PCa è una malattia molto elusiva, dal momento che i dati disponibili non consentono la discriminazione della forma indolente da quella aggressiva. Lo scenario attuale della gestione clinica di questi pazienti è quindi caratterizzato da “over-diagnosis” e “over-treatment” della forma indolente, e scarsa efficacia della terapia delle forme aggressive. Le implementazioni in questo campo sono dirette ad una migliore gestione clinica dei pazienti maschi urologici ed in particolare dei pazienti affetti da PCa. Si calcola che nel 2008 ci siano stati 382.000 nuovi casi di PCa nella sola Europa. Come UdR INBB, sono state identificate e validate una “gene signature” con potere diagnostico e prognostico del PCa in grado di identificare e discriminare la forma aggressiva e misurare la risposta personalizzata ai trattamenti terapeutici; è stata messa a punto una strategia terapeutica chemio-preventiva che ha portato alla riduzione della diagnosi di PCa del 90% in soggetti a rischio in assenza di effetti collaterali rilevanti, con trial clinico di fase II;
