The hypothesis is that leukemia cells loaded with TLR ligands are capable of activating the recently discovered helper function of NK cells, so these NK cells can effectuate the polarization of immature DCs to T helper type 1 (Th1)-polarised DCs (DC1NK). This cross-talk between the innate and adaptive immune system in which DC1NK play a central role, would then facilitate the activation of antigen-specific Th1 cells and cytotoxic T lymphocytes.

In dit project zullen we, in een model van acute myeloïde leukemie (AML), de invloed nagaan van Toll-like receptor (TLR)-ligand-geladen leukemische tumorcellen op de activatie van dendritische cellen (DCs) en natural killer (NK) cellen. De hypothese is dat TLR-ligand-geladen leukemiecellen in staat zijn om de recent ontdekte 'helper' functie van NK cellen te activeren, zodat deze op hun beurt de polarisatie van immature DCs tot T helper type 1 (Th1)-gepolariseerde DCs kunnen bewerkstelligen (DC1NK). Deze wisselwerking tussen het aangeboren en adaptieve immuunsysteem waarin DC1NK centraal staan, zou dan de activatie van antigeen-specifieke Th1 cellen en cytotoxische T lymfocyten (CTL) faciliteren.
