脳はいかようにも変化する”可塑性”を持つ組織です。脳の神経細胞が繰り返し刺激を受けると新たなシナプスが形成されて、情報伝達の効率が変化します。つまり、シナプス形成はまさしく学習の工事現場と言えるのです。では、ここではいったいどのような分子が働いているのでしょう？過去の研究で、神経伝達物質のセンソリン（sensorin）というタンパク質のメッセンジャーRNA(mRNA)は、核周辺に留まらずシナプス終末にまで運ばれ濃縮されていて、神経細胞が入力刺激を受けるとタンパク質に翻訳されることがわかっていました。その翻訳を阻害すると、シナプス形成が起こらないことも確認されました。このことから、センソリンのメッセンジャーRNAの神経末端への移動と翻訳がシナプス形成に重要であるといえます。 ここまでわかると、次の疑問が湧いてきました。どうやってmRNAが核からシナプス終末までの遠距離を移動しているのか？どうやってシナプス終末で翻訳が開始されるのか？そして、センソリンはどのようなメカニズムでシナプス形成に働くのか？これらの分子メカニズムを追求するために、私たちはイメージング技術を駆使して、研究を行っています。 まず、センソリンのmRNAの微細な局在を観察するために、蛍光in situ ハイブリダイゼーション法（FISH法）というイメージング技術を取り入れ、mRNAそのものを蛍光物質で標識し観察を行いました。さらにmRNAの様々な断片を作って観察を繰り返したところ、センソリンのmRNAがシナプス終末に移動するには、5’側のUTRにある66塩基が重要であるというところまで突き止めることができました。
