The light-sensitive photoreceptors made by the rod and cone cells in the retina also belong to the GPCR class. Moreover, they are the only GPCRs that are intrinsically sensitive to light. Functional damage to these photoreceptors, or pathological loss of the cells that bear them, results in inability to register light impinging on the retina – and is responsible for various types of visual impairment and certain forms of congenital blindness. In collaboration with researchers at the University of California in Berkeley, Trauner and his group have, for the first time, succeeded in restoring visual function to mice in which retinal photoreceptive cells had degenerated. The trick was to use a new synthetic switch to confer light sensitivity on the retinal ganglion cells in these mice, which normally respond to signals from the rods and cones upstream of them.

“Our methods are best suited for analysing the stoichiometry of proteins,” said Ulbrich, pointing out that their work involves a lot of counting. In future, the Freiburg researchers hope to find out the proportion of different subunits in a specific GPCR type and whether this proportion is absolutely necessary for the function of a receptor. In another project, the researchers plan to measure how many modulatory proteins can bind to an AMPA-type glutamate receptor while it is exerting its function. Biochemical and genetic methods cannot be used to count the number of proteins as they only show whether, and not how many, modulatory proteins are present.

Pure mouse ES-derived cardiomyocytes (CorAt®) are available in unlimited amounts, can be shipped frozen overnight, and are quality controlled showing no lot-to-lot variation. Protocols and applications include but are not limited to cardiac specific cytotoxicity, electrophysiology, GPCR functionality, siRNA transfection and hypertrophy induction. The cells express and use all essential ion channels and K, Na, and Ca currents have been recorded in patch clamp experiments. In addition, the expected effects of known ion channel modulators and cardio toxic substances are observed. Measurements have been validated on most instruments available.

Monoaminergic receptors including a number of adrenergic and dopaminergic subtypes are known as target proteins for CNS- and cardiovascular-active drugs and serve as model systems for understanding the structure, cell biology, and physiology of GPCR-ligand complexes. Brian Koblika’s lab at Stanford has developed biophysical methodology to monitor the structure and ligand-induced conformational changes of GPCRs. Peter Gmeiner’s group in Erlangen has substantial experience in the design and organic synthesis of novel, highly specific GPCR agonists and antagonists. The collaborative investigations resulted in a crystal structure of the first agonist-bound GPCR. Using crystallography as a tool to develop medicinal chemistry of a given GPCR, collaboration between the Bavarian and the Californian research group will lead to novel insights into structure activity relationships that will be transferred to related receptor ligand systems.
