In 2005, researchers from the University of Saskatchewan in Saskatoon, Canada, turned the age-old claim that weed kills brain cells upside down. They found that the administration of synthetic cannabinoids (designed to mimic the effects of THC and other natural cannabinoids) could actually stimulate brain cell growth.

The Pfizer Research Prize 2017 in oncology basic research goes to Dr. Christian Schürch (Institute of Pathology) and PD Dr. Carsten Riether (Department of Clinical Research). They found that the CD27 receptor plays an important role for the survival and proliferation of leukemia stem cells (LSCs) in chronic myeloid leukemia (CML). Tyrosine kinase inhibitors (TKIs), the current standard of care for CML, lead to up-regulation of the CD27-ligand (CD70) on LSCs which induces therapy resistance by activating the Wnt signaling pathway. Combination therapy of TKI and anti-CD70 antibodies overcomes this resistance mechanism, eliminates LSCs and cures CML in mouse models.

For four years the colonists managed to survive. Albion was tough on them and took its toll as they learned which things were safe to eat and which might eat them. Divisions soon emerged and the group splintered off into various sub-factions. The largest of these was led by a man called Gibson. A butcher by trade, with a good head for figures, Gibson had got on the wrong side of the King due to his outspoken views on taxation. He was a methodical man, who knew how to divide things fairly and wasn’t afraid to speak out. He was also handy with a meat cleaver.

The experiments were carried out at the Max Born Institute at a 12 meter long high-harmonic generation (HHG) beamline. "The observation that argon clusters were strongly ionized even at moderate NIR laser intensities was very surprising", explains Dr. Bernd Schütte from MBI, who conceived and performed the experiments. "Even though the additional XUV laser pulse is weak, its presence is crucial: without the XUV ignition pulse, the nanoparticles remained unaffected and transparent for the NIR light (Figure 2)." Theorists around Prof. Thomas Fennel from the University of Rostock modelled the light-matter processes with numerical simulations and uncovered the origin of the observed synergy of the two laser pulses. They found that only a few seed electrons created by the ionizing radiation of the XUV pulse are sufficient to start a process similar to a snow avalanche in the mountains. The seed electrons are heated in the NIR laser light and kick out even more electrons. "In this avalanching process, the number of free electrons in the nanoparticle increases exponentially", explains Prof. Fennel. "Eventually, the nanoscale plasma in the particles can be heated so strongly that highly charged ions are created."

Learning and memory relies on the proper processing of signals that stimulate neuronal cells within the brain. Prof. Jean Pieters’ team at the Biozentrum of the University of Basel, together with an international team of scientists, has uncovered an important role for the protein coronin 1 in cognition and behavior. They found that a lack of coronin 1 in mouse and in man is associated with poor memory, defective learning and aggressive behavior. The results, recently published in “PLOS Biology”, identify a novel risk factor for neurobehavioral dysfunction and reveal a molecular pathway involved in transferring information within neurons.

Understanding the connection between structure, regulation and function of the Sip1 proteins was possible only as a result of the close collaboration between scientists from different research areas at the Department of Chemistry. In biological and biochemical experiments they first determined the significance of Sip1 for the survival of nematode embryos. They discovered how the heat shock protein prevents clotting of important embryonic proteins during heat stress when activated by a low pH value.

Among these substances is HC-toxin, which has now been investigated by researchers of the Clinical Cooperation Unit "Pediatric Oncology" at the German Cancer Research Center (Deutsches Krebsforschungszentrum, DKFZ). They found out that neuroblastoma cells lose several of their cancer-typical properties when under the influence of the substance: They divide less frequently, show less invasive growth and even their outside appearance resembles healthy nerve cells again. These effects were observed to be stronger than with other HDAC inhibitors investigated previously.

Xiaonan Han, Richard Moriggl and their team of researchers from the Cincinnati Children´s Hospital in collaboration with several groups from the University of Veterinary Medicine, the Medical University Vienna and the Ludwig Boltzmann Institute for Cancer Research, now link STAT5 signalling to IESC replenishment upon injury. Using genetic model systems, the international consortium associates depletion of Stat5 gene to decreased proliferation of a specific subset of IESC pools. Mice lacking STAT5 were more prone to gastrointestinal injury upon colitis, infectious disease or gamma-irradiation challenge. Inducible expression of STAT5 increased IESC proliferation, accelerated crypt regeneration, and conferred resistance to intestinal injury. Furthermore, ectopic activation of STAT5 in mouse or human stem cells promoted self-renewal. Therefore STAT5 is a functional therapeutic target to improve the IESC regenerative response to gut injury.

The mirror carp is a popular fish, not just with anglers. Cooks also appreciate it for its almost entire lack of scales, which makes it considerably easier to prepare than other types of fish. This “user-friendly” characteristic harks back to medieval monks who bred them in their monasteries. Several centuries later, a group of scientists working at the Max Planck Institute for Developmental Biology in Tübingen have now uncovered the secret behind this characteristic of this breed. They discovered that a mutation in one single gene can prevent the formation of scales. This gene codes for the fibroblast growth factor receptor 1. What amazed the biologists, however, was the fact that, despite this, the fish remains viable. This receptor is, after all, involved in many important processes relating to embryonic development. Thus, a mutation should therefore actually cause serious problems. Mice embryos with defects in this gene, for instance, do not develop. This gene is also essential for survival in humans. If its function is partially impaired, it gives rise to a genetic disorder known as Kallmann syndrome, which is associated with a decreased ability to smell as well as deficiencies of the testes or ovaries.

As I was investigating the possibilities of performing under Dresden bridges, Mariana from our Divus studio called me. She had just crashed the car borrowed from DJ BLN when distributing the magazine, and so yet another car was wrecked. Since then, we have had no car at Divus, and we don’t want any. The hotel next to our studio and editorial office took up all the parking places anyway, so what for anyway. We bought big shopping bags on wheels.

Off the coast of Iceland, over three hundred feet below the surface of the ocean, hot volcanic rock began to seep into the cold ocean water in 1963. By 1967, over half a cubic mile of new, hot rock had formed a brand new island where there was once only ocean. This new island was named Surtsey. The following summer, scientists arrived on the island to study it. They found that a basalt cliff on the beach had been eroded over 300 feet inland by the sea in less than five years. Several parts of the island already had terraced beaches up to 300 feet wide. On the north side of the island, newly formed sand had formed a sand bar which extended almost a thousand feet out from the original island.

In a joint study international scientists investigated how clouds and hence rain develops over the Amazon tropical rain forest. They found out that the rain is generating further rain by downdrafts entraining fine particles from the upper atmospheric layers to the layer where clouds are formed. As, in this area, there are no cloud condensation particles from anthropogenic pollution, these findings are important to help understanding how precipitation developed in preindustrial times.

The word ‘carat’, used to denote the weight of gold and diamonds, can be traced back to Ancient Greek as well. They discovered that the seeds of the carob tree had a constant weight of about 0.2 milligrams. As a result, the seed, called ‘keration’ in Greek, was used as a weight unit.

In earlier work, Pieters’ team discovered the protein coronin 1 as being essential for the proper transduction of signals in immune cells. In mice lacking coronin 1 the researchers further investigated the molecular mechanism. Surprisingly, these mice showed aberrant behavior. In particular, mice lacking coronin 1 appeared to be far more aggressive and display extreme grooming activity, an indication of reduced sociability. An in-depth analysis in collaboration with scientists from the Friedrich Miescher Institute in Basel and the University of Bordeaux unveiled profound learning and behavioral problems and severe defects in the ability to activate neurons in the absence of coronin 1.

The second experiment is a fascinating experiment. It's a military experiment. What they did in essence, was they took some human DNA, some scrapings from the tissue of inside the mouth of a donor, or volunteer. And they placed this DNA in a device that could measure it's effects in one room of a building, while the donor that the DNA came from is in another room in the same building.

The Bindu Chakra bestows physical and mental health, vitality and youthfulness, because it produces “the nectar of immortality” (Amrita). This nectar usually drops into the Manipura Chakra where it is burned by the digestive fire without being fully utilised by the body. For this reason the Rishis of ancient times sought a method to collect this valuable nectar and discovered that the flow of nectar can be arrested with the help of the tongue and Vishuddhi Chakra. The tongue contains subtle energy centres, each corresponding to an organ or area of the body. In the Yoga techniques of Ujjayi Pranayama and Khechari Mudra, the tongue curbs the flow of nectar and stores it in the Vishuddhi Chakra. Like a homeopathic medicine it is then redistributed throughout the whole body via the subtle energy channels, where its healing effects unfold.

Investigations at the molecular level are insufficient in recognizing discomfort, stress, fear, pain and suffering – for example an increased concentration of stress hormones in the blood can indicate agitation in a positive sense. “Experiments must therefore be refined so as to discover an animal’s specific preferences and dislikes. And it is also important to understand what suffering means for different animals, particularly when their biological requirements and sensory abilities are very different to our own,” explains Weary, whose knowledge of such experimental set-ups is highly valued by Braithwaite and Droege. For instance, to better understand the motivation of dairy cow to access pasture, cows were trained to open a weighted gate. The maximum weight the cows were willing to push could then be compared to the weight they were willing to push when they were hungry. The study showed that cows would ‘work’ very hard to go outdoors, pushing about the same weight as they are willing to push to access fresh feed after their morning milking. “Nobody would deny cows their breakfast but they are often prevented from grazing in the pasture,” says Weary, thus throwing into question a present-day practice on many farms. In other work Weary and his students have developed methods of assessing mood states in animals by testing their responses to ambiguous stimuli. They have found that dairy calves will show a negative judgment bias (i.e. treat a neutral stimulus as if it was negative) when experiencing the pain in the hours after dehorning (a common farm procedure). Interestingly, calves showed a very similar judgment bias in the hours after separation from their mother at weaning, suggesting some similarity in the effects of emotional and physical pain on mood in animals.

In a new study, Sebastian Bathiany and his colleagues Dirk Notz, Thorsten Mauritsen, Victor Brovkin and Gaby Raedel from MPI-M have examined the loss of Arctic sea-ice. They found that abrupt sea-ice loss can occur in winter even without feedback processes. The large sensitivity of the winter sea-ice area is caused by the asymmetry between melting and freezing. The scientists also addressed whether abrupt winter ice loss might be a reversible phenomenon and whether it could be anticipated from observations.

Years ago, free software developers noticed this discomfort reaction, and some started exploring an approach for avoiding it. They figured that by keeping quiet about ethics and freedom, and talking only about the immediate practical benefits of certain free software, they might be able to “sell” the software more effectively to certain users, especially business. The term “open source” is offered as a way of doing more of this—a way to be “more acceptable to business.” The views and values of the Open Source movement stem from this decision.

Using clinical data from solid tumor patients, the researchers evaluate the potential and limitation of various targeted combination treatments. They find that even if there is only one possible mutation in the genome that has the potential to cause resistance to all administered drugs, combination therapy will not lead to long-term disease control in most patients. However, if no possible cross-resistance mutations exist, and full resistance can only be gained if a cell has sequentially received mutations where each makes it resistant to one of the administered drugs, dual therapy can result in long-term disease control for the vast majority of patients. Triple therapy will then only be needed for patients with very large tumor burden. Furthermore, the researchers demonstrate that the current practice of administering targeted agents sequentially is a recipe for certain treatment failure and that simultaneous administration of targeted agents is essential for long-term disease control. Even when there are potential mutations conferring cross-resistance, simultaneous use of targeted drugs offers some hope for cure where sequential therapy fails with certainty. This research thus realizes the advantages of simultaneous combination therapy in solid cancer treatment, with a hope to stimulate efforts to combine agents much earlier in the drug development process.
