Yes, Celepuk Reban (Otus lempiji) is one of the owl species widely found in Yogyakarta, particularly in rural areas like Pakem, where we "hunted" at New Year's Eve several years ago. We intentionally wandered in the middle of the night around the village, all to just "hunt" for nocturnal animals-not to be caught and kept in a cage, but only to see them in their natural environment.

Ya, si Celepuk Reban atau nama latinnya Otus lempiji merupakan salah satu burung hantu yang umum dijumpai di Yogyakarta-khususnya di daerah pedesaan seperti Pakem, tempat 'berburu' kami pada malam tahun baru beberapa tahun yang lalu. Kami sengaja berkeliaran malam-malam di sekitar desa hanya untuk 'berburu' hewan-hewan malam yang bisa kami temui-bukan untuk ditangkap, melainkan hanya untuk melihatnya secara lansung di alam bebas. Acara sederhana ini memang sengaja kami adakan untuk mengisi malam tahun baru dengan cara yang berbeda, bukan hanya berpesta pora seperti yang dilakukan oleh kebanyakan orang pada saat itu.

Lucie Zinger, Antje Boetius und Alban Ramette vom Bremer Max-Planck-Institut für Marine Mikrobiologie haben in Kooperation mit ihren Kollegen aus mehreren amerikanischen Forschungsinstituten die bakterielle Diversität in mehr als 500 Proben aus allen Teilen der Weltmeere untersucht. Darunter waren verschiedene Proben aus der Wassersäule (dem pelagischen Bereich), sowie aus der Bodenzone des Meeres (dem benthischen Bereich). Die Proben kamen sowohl aus küstennahen Gewässern als auch aus der Tiefsee, aus sauerstofffreien Gewässern und Sedimenten und von Hydrothermalquellen. Die Wissenschaftler extrahierten die in den Proben befindliche Erbsubstanz (DNA) und sequenzierten daraufhin einen stark variablen Bereich des bakteriellen ribosomalen RNA-Gens, den V6–Bereich, in einem besonderen Verfahren, das einen sehr hohen Durchsatz erlaubt. Nachdem sie die einzelnen Sequenzen zu mehr als 120.000 OTUs (operational taxonomic units, d.h. zu taxonomischen Einheiten) zusammengesetzt hatten, nutzten sie multivariate Datenanalysen, um herauszufinden, inwiefern die einzelnen Faktoren wie Umwelt, Geographie und Primärproduktion (also die Produktion von Biomasse im Wasser) die Veränderung der mikrobiellen Gemeinschaften beeinflussen.

The rumen bacterial community was fractionated and analyzed as three separate fractions: populations in the planktonic, loosely attached to rumen digesta particles, and tightly attached to rumen digesta particles. Six hundred and thirteen operational taxonomic units (OTUs) belonging to 32 genera, 19 families, and nine phyla of the domain Bacteria were identified from 1014 sequenced clones.

Nous avons étudié, à l’aide d’analyses de la banque de clones du gène de l’ARNr 16S, la composition, la répartition et la biodiversité des espèces des communautés bactériennes dans le rumen de vaches ayant reçu de la luzerne ou du triticale. La communauté bactérienne ruminale a été divisée et analysée en trois fractions : la population planctonique, la population de bactéries fixées de manière lâche aux particules du digesta ruminal et la population de bactéries fixées fermement à ces mêmes particules. Nous avons identifié, d’après 1 014 clones séquencés, 613 unités taxonomiques opérationnelles (UTO) appartenant à 32 genres, 19 familles et 9 phylums du domaine des bactéries. De ces 613 UTO, 451 étaient de nouvelles espèces. Ces séquences bactériennes étaient réparties de façon différente entre les trois fractions dans le digesta ruminal des vaches ayant reçu de la luzerne ou du triticale. L’estimation de Chao 1 a révélé que, dans les deux communautés, la population de bactéries fixées fermement aux particules était plus diversifiée que la population planctonique et que la population de bactéries fixées de manière lâche aux particules. Le programme S‑Libshuff a permis de déceler des différences significatives dans la composition entre deux fractions, quelles qu’elles soient, dans le rumen des vaches ayant reçu la même ration et entre les communautés bactériennes des vaches ayant reçu les rations de luzerne et de triticale. La richesse des espèces bactériennes chez les vaches ayant reçu de la luzerne et du triticale a été estimée à 1 027 et à 662, respectivement. La diversité de la communauté bactérienne ruminale examinée dans la présente étude est plus grande que ce que les études antérieures ont démontré, et les différences dans la composition de la communauté entre deux rations à teneur élevée en fibres ont des effets sur la sélection des échantillons pour les applications en aval de la métagénomique.
