The researchers encountered a gene that could be an important determinant for the gender-specific differences in Alzheimer’s disease pathology. The gene, called ubiquitin-specific peptidase 9 (USP9), influences the activity of another gene that encodes the microtubule associated protein tau (MAPT).
Die Forscher stießen auf ein Gen, das als wichtiger Einflussfaktor für die geschlechtsspezifischen Unterschiede in Bezug auf Alzheimer infrage kommt. Das Gen namens Ubiquitin-spezifische Peptidase 9 (USP9) beeinflusst die Aktivität eines Gens, welches das Tau-Protein kodiert (Mikrotubuli-assoziertes Protein Tau, MAPT). Mutations in 3 genes are responsible for most of the FTD genetic cases: microtubule associated protein tau (MAPT), progranulin (PGRN) and chromosome 9 open reading frame 72 (C9orf72). Genetic or sporadic FTDs share common neuropathological features such as neuronal Tubulin-Associated Unit (TAU), Tar-DNA binding Protein 43 (TDP43), or Fused in Sarcoma (FUS) inclusions.
Up to 50% of ALS patients develop FTD symptoms and around 15% of FTD patients display motor neuron dysfunction typical of ALS. To date, no treatment is available for these disorders, and the molecular mechanisms at stake in the different pathological subtypes remain elusive.

So far only mutations in the microtubule associated protein tau gene (MAPT) on chromosome 17q21 have been identified for FTD linked to chromosome 17, with a frequency of 10 tot 43% in familial FTD.
Frontotemporale dementie (FTD) is de tweede meest frequente vorm van neurodegeneratieve dementie in mensen jonger dan 65 jaar. Tot 50% van de FTD patiënten hebben een positieve familie historiek voor dementie en in de meerderheid van de families wordt de ziekte op autosomaal dominante wijze overgeërfd. Tot nu toe werd alleen mutaties in het microtubuli geassocieerde proteïne tau (MAPT) gen op chromosoom 17q21 geïdentificeerd als causaal voor FTD gekoppeld aan chromosoom 17, met een frequentie van 10 tot 43% in familiale FTD. In een aantal autosomaal dominante FTD families met 17q21 koppeling werden geen MAPT mutaties gedetecteerd (FTDU-17). Met het voorgestelde project willen we verder onderzoek doen naar het genetisch defect bij FTDU-17. We zullen de strategie volgen van positionele klonering in autosomaal dominante FTD families.
